Overview
Thymosin Beta-4 is a 43-amino-acid peptide encoded by the TMSB4X gene and the most abundant member of the beta-thymosin family. It has the molecular formula C212H350N56O78S, a molecular weight of approximately 4963 g/mol, and CAS number 77591-33-4. The peptide was first isolated from calf thymus tissue in 1981 and has since been studied as a principal G-actin–sequestering protein in mammalian cells.
Structurally, Thymosin Beta-4 carries an N-terminal acetyl group on its serine residue. Its full sequence is Ac-Ser-Asp-Lys-Pro-Asp-Met-Ala-Glu-Ile-Glu-Lys-Phe-Asp-Lys-Ser-Lys-Leu-Lys-Lys-Thr-Glu-Thr-Gln-Glu-Lys-Asn-Pro-Leu-Pro-Ser-Lys-Glu-Thr-Ile-Glu-Gln-Glu-Lys-Gln-Ala-Gly-Glu-Ser. The central LKKTETQ motif is the defining actin-binding domain and has been the focus of in-vitro biochemical characterization.
